Abstract


Scanning electron microscopic characteristics of osteoclasts in situ preserved by enzymatic digestion method were studied. The calvariae of albino rat of 6 days of age were asceptically removed and cultured in incubator for 3 days. PTH and PGE2
were
added to the culture media and cultured in incubator for 3 days with the calvariae. Normal PTH-treated and PGE2-treated calvariae were diagested with enzyme solution containing 0.1% collagenase, 0.1% collagenase, 0.05% trypsin and 4m M EDTA for
20
min,
4 times. The osteoclasts in situ were processed for the scanning electron microscopic observation. Also the ventral surface of the osteoclast was exposed by turning a cell inside out through attaching the dorsal surface of osteoclast on adhesive
tape
after SEM observation of cultivated cells. Exposed underside of osteoclasts on the tape were examined by SEM.
@ES The results were as follows :
@EN 1. The contour of osteoclasts was variable. The dorsal surface of the osteoclast was rough with prominent microvilli and leaf-like processes. In some osteoclasts, the surface was smooth, or peripheral fillopodia were observed.
2. The functional surface of osteoclasts were composed of centrally located ruffled and marginally located non-ruffled ones.
3. PTH and PGF2 did not affect the surface morphology of the normal osteoclast.
4. In PTH and PGE2 group, osteoclasts were generally enlarged. Fillopodia of the osteoclast were more prominent in PGE2 group than PTH group.
